چکیده

CCNE1 is one of the most frequent gene amplifications in high grade serous ovarian cancers (HGSOCs), which occurs in at least 20% of HGSOC and in 30% of established ovarian cancer cell lines. Studies with both primary and metastatic ovary tumor specimens further suggest that the abundance of CCNE1 protein expression correlates with tumor progression and predicts a poor prognosis in ovarian cancer patients[1]. It is noted that, CCNE1 is critical for the growth of ovarian cancer cell lines with elevated CCNE1 expression but not cells without CCNE1 overexpression [1]. Furthermore, CCNE1 gene amplification-associated CCNE1 overexpression has been linked to the development of chemo-resistance in ovarian cancer[1]. Thus, accumulated findings implicate that CCNE1 may be a promising therapeutic target for ovary tumors with elevated CCNE1 expression. However, developing small molecules to target CCNE1 directly is unlikely, because CCNE1 acts as a regulatory subunit of cyclindependent kinase (Cdk) complex rather than as an enzyme or receptor. It is well known that CCNE1 mainly coordinates with Cdk2 to facilitate G1/S progression of cell cycle. In ovary tumors, elevated CCNE1 level is often correlated with higher Cdk2 expression and most of CCNE1associated tumor promoting effects require the participation of Cdk2. Thus, targeting Cdk2 may be an attractive alternative given the current availability of small molecule Cdk2 inhibitors. SNS-032 (BMS387032) is a selective inhibitor of CDK2, and has been evaluated in Phase I study for patients with either chronic lymphocytic leukemia or multiple myeloma, as well as clinical safety assessment for the treatment of select advanced solid tumors. We showed that ovarian cancer cells with elevated CCNE1 expression are at least 40 times more sensitive to SNS-032 than those without CCNE1 overexpression. Moreover, we demonstrated that SNS-032 effectively suppresses the tumorigenicity of ovarian cancer cells by prolonging the survival of animals bearing tumors derived from ovarian cancer cells with elevated CCNE1 expression and inhibiting peritoneal metastatic colonization. These results suggest that ovary tumors with elevated CCNE1 expression may be staged for Cdk2-targeted therapy. Editorial

Cell cycle regulation and anticancer drug discovery

Cellular growth, development, and differentiation are tightly controlled by a conserved biological mechanism: the cell cycle. This cycle is primarily regulated by cyclin-dependent kinase (CDK)-cyclin complexes, checkpoint kinases, and CDK inhibitors. Deregulation of the cell cycle is a hallmark of the transformation of normal cells into tumor cells. Given its importance in tumorigenesis, severa...

Cyclin E1 Inhibition can Overcome Sorafenib Resistance in Hepatocellular Carcinoma Cells Through Mcl-1 Suppression.

PURPOSE To clarify the effects of cyclin E1 suppression on antitumor efficacy of sorafenib in hepatocellular carcinoma cells and to explore the potential of combining sorafenib with cyclin-dependent kinase (CDK) inhibition in therapy. EXPERIMENTAL DESIGN The effects of cyclin E1 suppression on sorafenib-induced apoptosis were tested in both sorafenib-sensitive (Huh-7 and HepG2, IC50 5-6 μmol/...

Molecular Pathways Molecular Pathways: CDK4 Inhibitors for Cancer Therapy

Unrestrained growth is the hallmark of cancer, and disrupted cell-cycle regulation is, therefore, common. CDK4 is the key regulator of the G1–S transition. In complex with cyclin D, CDK4 phosphorylates retinoblastoma protein (Rb) and drives cell-cycle progression, a process inhibited by p16. The p16– CDK4–cyclin D–Rb is aberrant in themajority of cancers and is, thus, a logical target for antic...

Anticancer effects of an extract from the scallop Patinopecten yessoensis on MCF-7 human breast carcinoma cells

Patinopecten yessoensis, is a species of scallop and a marine bivalve mollusk. In traditional East Asian medicine, scallop meat is used as a drug for the treatment of diabetes, pollakisuria, and indigestion. The present study was conducted in order to examine the potential anticancer effects of scallop flesh extract (SE) on MCF-7 human breast cancer cells. An MTT assay was used to evaluate cell...

Proteasome-mediated destruction of the cyclin a/cyclin-dependent kinase 2 complex suppresses tumor cell growth in vitro and in vivo.

Cyclin-dependent kinases (cdks) represent potentially promising molecular targets for cancer therapeutic strategies. To evaluate the antitumor activity of selective cyclin/cdk inhibition, we constructed a chimeric protein composed of a F-box protein (TrCP) fused to a peptide comprising the cyclin/cdk2 binding motif in p21-like cdk inhibitors (TrCP-LFG). We now demonstrate that endogenous cyclin...
